Functional non-animal biobarrier models in research and risk assessment

Overall goal of the workshop:This workshop aims to bring together experts in the field of biobarriers for extensive exchange of knowledge, definition of the status quo, identification of shortcomings and pitfalls as well as addressing relevant questions in this field.

Abstract:

The biological barriers –biobarriers- of the human body prevent and control the entrance of microorganisms, substances and particles into the system and organs. The skin poses an external biobarrier, endothelium, blood-brain-barrier (BBB), blood-cerebrospinal fluid barrier (BCB), brain parenchyma, the gastrointestinal and mouth mucosa, the respiratory system and the placenta pose internal biobarriers.

Pharmaceuticals, chemicals, cosmetics and food ingredients take effect dependant on their distribution on the cellular, organ and the whole system level. Some compounds are to trespass the biobarrier by diffusion or active transport, e.g. therapeutics and nutrition. Indeed, there is growing demand for development of carrier molecules to enable the barrier translocation of particular pharmaceuticals to sites where they are to be delivered. The active or passive crossing of other compounds may pose a threat, e.g. the translocation of distinct nanoparticles through the placental biobarrier may impede embryogenesis, and thus is undesirable. In addition the biobarrier itself may be prone to toxic and other adverse effects of compounds they are exposed to, leading to compromised coherence of the barrier.

For examination and evaluation of transport and diffusion of substances, experimental carrier molecules and particles through different biobarriers and their effects on the various barriers in vitro, ex vivo and in silico systems are already established. These models are to be discussed in this workshop with regard to their functionality and fit-for-purpose from academic, industrial and regulatory point of view.
